    Resumen

    Los materiales plásticos son uno de los materiales más utilizados en la industria alimentaria para el envasado de alimentos. Los materiales poliméricos derivados del petróleo son los más utilizados, debido a que presentan numerosas ventajas como su disponibilidad a gran escala, relativo bajo costo de producción, gran versatilidad y relativamente buenas propiedades de barrera. Sin embargo provienen de una fuente de recursos no renovable. Por otro lado existen interacciones entre los envases plásticos y los alimentos que genera ciertas restricciones de uso de algunos polímeros, pero estas interacciones se pueden aprovechar para el desarrollo de envases activos. Resulta de gran interés estudiar el desarrollo de nuevos materiales plásticos para aplicaciones en envases, empleando materiales de origen renovable, de menor impacto ambiental. Una alternativa es el PHB (polihidroxibutirato), que además de ser biodegradable, es un polímero obtenido a partir de fuentes renovables. Sin embargo este polímero presenta algunas desventajas, como por ejemplo una baja estabilidad térmica, baja deformación a la rotura, alta rigidez y fragilidad. El objetivo principal del presente trabajo de tesis es el desarrollo de formulaciones a partir de PHB, con incorporación de nanorefuerzos (montomorillonita natural y montmorillonitas orgánicamente modificadas) e incorporación de compuestos activos, de manera de lograr; por un lado; mejoras en las propiedades físico-mecánicas, de barrera y resistencia a la humedad, limitaciones naturales de biopolímeros y polímeros biodegradables; y por otro lado; materiales que presenten actividad antimicrobiana.
    Se obtuvieron y se estudiaron compuestos de PHB (matriz polimérica) con distintos tipos de montmorillonitas comerciales (montmorillonita natural Cloisite® Na y orgánicamente modificadas Cloisite® 30B, 93A, 10A, 15A y 20A) con el objetivo de estudiar la influencia de la incorporación de cada una de estas arcillas en las propiedades de la matriz polimérica. Las mezclas se realizaron con 3% de montmorillonita y 0,2% de irgafos 168, un antioxidante secundario que se utilizó como extendedor de cadena durante el procesamiento. Por otro lado se obtuvieron y se evaluaron compuestos de PHB (matriz polimérica) y una montmorillonita natural (Cloisite® Na) intercambiada con cationes Cu++, con el objetivo de estudiar la actividad antimicrobiana del material obtenido. Todos los compuestos se obtuvieron utilizando el método de intercalación en estado fundido en un mezclador discontinuo con un tiempo de mezcla de 10 minutos y a una temperatura de mezclado de 165°C. Se estudió la estructura y morfología de los materiales obtenidos a través de DRX, reología y SEM; y el comportamiento térmico por DSC y MOP. Además se realizó la medición del peso molecular de los materiales por GPC, se midieron propiedades mecánicas y se midió la permeabilidad al vapor de agua siguiendo la norma ASTM E96 / E96M – 05.
    Los resultados mostraron una caída en el peso molecular de la matriz PHB en las condiciones de procesamiento estudiadas. La incorporación de un agente extendedor de cadena no tuvo impacto en las propiedades de la matriz. Con la incorporación de arcillas orgánicamente modificadas Clo 30B, 10A, 15A y 20A se obtuvieron nanocompuestos con un mayor grado de intercalación/exfoliación, pero al mismo tiempo la degradación del PHB se vio acelerada, dando lugar a una caída drástica en el peso molecular de los materiales obtenidos con estas arcillas. Se observó una disminución en la temperatura de fusión y valores similares de temperatura de cristalización desde el fundido, indicando que estas arcillas no actuaron como agentes nucleantes durante la cristalización del PHB. La incorporación de arcilla orgánicamente modificada Clo 93A permitió obtener nanocompuestos con mejores propiedades respecto de la matriz de PHB. Se obtuvieron compuestos de PHB y Clo 93A en dos porcentajes distintos, 3 y 5 % de arcilla, realizando las mezclas con y sin el extendedor de cadena. Se comprobó la existencia de un cierto grado de intercalación/exfoliación mediante DRX y reología para los compuestos realizados con 3 y 5% de arcilla (con y sin el extendedor de cadena). No se observaron cambios significativos entre los compuestos que tienen irgafos respecto a los que no tienen (para un mismo porcentaje de arcilla). La incorporación de Clo 93A no produjo cambios significativos en la temperatura de fusión del PHB. El compuesto realizado con 5% de arcilla y con 0,2% de irgafos presentó un aumento significativo en el porcentaje de cristalinidad (77% respecto a 55, 8% para el PHB). Por otro lado, se observó un aumento en la temperatura de cristalización desde el fundido para todos los compuestos, aunque el incremento no fue significativo. No se observaron diferencias significativas en el módulo tensil de los nanocompuestos obtenidos a partir del ensayo de tracción, indicando que el grado de intercalación/exfoliación alcanzado no fue suficiente para que la arcilla actuara como refuerzo de la matriz polimérica. Además los compuestos realizados con 5% de arcilla (con y sin irgafos) mostraron una caída en los valores de tensión a la carga máxima, mientras que los compuestos realizados con 3% alcanzaron valores similares a los de PHB. Con el agregado de 5% de arcilla se observó una caída en la permeabilidad al vapor de agua.
    Por último, los resultados de la evaluación de la actividad antimicrobiana, realizada a mezclas de PHB y montmorillonita natural Clo Na intercambiada con cationes Cu++, mostraron que los materiales obtenidos presentaron actividad frente a S. aureus (utilizado como modelo de microorganismo Gram positivo) y E. coli (utilizado como modelo de microorganismo Gram negativo).

    Biodegradable polymers with antimicrobial activity

    Abstract

    Plastics are one of the materials most commonly used in the food industry for food packaging. Petroleum polymeric materials are the most commonly used, since they have numerous advantages as their large scale availability, low cost of production, versatility and good barrier properties. However they come from a non-renewable source. On the other hand there are interactions between food and plastic containers that generate certain restrictions on the use of certain polymers, but these interactions can be leveraged for the development of active packaging. It is of great interest to study the development of new plastic materials for packaging applications, using materials from renewable sources, with less environmental impact. A possible alternative is the PHB (polyhydroxybutyrate), that beside the fact it is biodegradable, it is also a polymer obtained from renewable sources. However this polymer has some disadvantages, such as low thermal stability, low strain at break, high rigidity and brittleness. The present thesis main objective is to develop formulations of PHB with the incorporation of nano-reinforcement (natural and modified montmorillonites, and modified montmorillonites with active compounds) in order to guarantee improvements, on one hand, on the physical-mechanical, barrier properties and moisture resistance, natural limitations of biodegradable polymers and biopolymers, and on the other hand, to produce materials that have antimicrobial activity).
    Compounds of PHB (polymer matrix) with different types of commercial montmorillonites (natural montmorillonite Cloisite® Na and Cloisite® 30B, 93A, 10A, 15A and 20A organically modified) were collected and studied in order to analyze the influence of the incorporation of each of these clays on the polymer matrix properties. Mixtures were made with 3% of montmorillonite and 0.2% irgafos 168, which was used as a chain extender during processing. On the other hand, compounds of PHB (polymer matrix) and a natural montmorillonite (Cloisite® Na) exchanged with cations Cu++ were obtained and assessed, in order to analyze the antimicrobial activity of the obtained material. All compounds were obtained using the melt intercalation method in a batch mixer with a mixing time of 10 minutes and mixing at a temperature of 165 ° C. The structure and morphology of the materials obtained were studied through XRD, rheology and SEM; and the thermal behavior by DSC and MOP. It was also carried out the measurement of the molecular weight of materials by GPC, the measurement of mechanical properties and permeability to water vapour according to the standard ASTM E96 / E96M - 05)
    The results showed a drop in the molecular weight of the PHB matrix in the processing conditions studied. The incorporation of an extender chain agent did not have an impact on the properties of the matrix. With the addition of the organically modified clays, Clo 30B, 10A, 15A, and 20A, nanocomposites were obtained with a greater degree of intercalation/exfoliation, but at the same time the degradation of PHB was accelerated, resulting in a drastic drop in the molecular weight of the materials obtained with these clays. A reduction in the melting temperature and similar values for crystallization temperature were observed, indicating that these clays did not act as nucleating agents during the crystallization of the PHB. The incorporation of organically modified clay Clo 93A allowed to obtain nanocomposites with improved properties than the PHB matrix. Compounds of PHB and Clo 93A in two different percentages, 3 and 5% of clay were obtained, with and without the chain extender). It was proved the existence of a certain degree of intercalation/exfoliation using XRD and rheology of the compounds with 3-5% clay (with and without the chain extender). No significant changes among the compounds having irgafos were observed in relation to those without (with the same percentage of clay). The incorporation of Clo 93A did not produce significant changes in the melting temperature of the PHB. Compound made with 5% clay and 0.2% of irgafos showed a significant increase in the percentage of crystallinity (77% compared to 55, 8%) for the PHB. Furthermore, an increase was observed in the crystallization temperature from the molten for all compounds, although the increase was not significant. There were not significant differences in the tensile module of the nanocomposites obtained from the tensile test, indicating that intercalation/exfoliation grade reached was not enough for clay to act as a reinforcement of the polymer matrix. In addition compounds made with 5% clay (with and without irgafos) showed a drop in the voltage at maximum load, while compounds made with 3% reached similar values as PHB. With the addition of 5% of clay a drop in permeability to water vapor was observed.
    Finally, the results of the evaluation of the antimicrobial activity, carried out by mixtures of PHB and natural montmorillonite Clo Na exchanged with cations Cu++, showed that the materials obtained had activity against S. aureus (used as a Gram positive model organism) and e. coli (used as a Gram negative model organism).
